miRNA-mediated reprogramming. miRNAs achieve reprogramming potentially by repressing the repressors of pluripotency genes. Genes involved in cell cycle progression, epithelial-mesenchymal transition (EMT) and epigenetic regulation are among genes that are targeted by the miR-302/367 cluster, but there are probably multiple important targets yet to be determined. Inhibition of HDAC2 by VPA, in conjunction with the miRNAs, is likely to enable reprogramming by promoting the activation of pluripotency genes.
