Comparison of string-of-beads with and without spacer sequences. Average junction cleavage likelihood (a), recovery percentage (b), and neo-immunogenicity (c) were measured for optimal string-of-beads designs with, without, and fixed AAY spacers. The string-of-beads constructs comprised three to 30 randomly selected epitopes. For each set size, the sampling was repeated 50 times. The maximum spacer length was set to k = 6. Black error bars and colored outlines represent the 68 % confidence intervals
