Overexpression of PACS1 resulted in an altered density of dendritic spines. Representative pictures of cortical neurons transfected with control (pCDH-GFP empty) and PACS1 overexpression vectors at DIV14. Only the GFP channel is shown to outline dendrite morphology (scale bar 10 μm). Branches of dendrites were imaged in each corresponding neuron (scale bar 5 μm). A two-tailed t test was used to determine whether the difference in dendritic spine density was statistically significant. To quantify the density of each dendritic spine subtype, 2-way ANOVA with multiple comparison testing using the Bonferroni correction (P = 0.0047) was used. More than 40 dendrites from 20 neurons were analysed in each group (control or PACS1 overexpression). The error bars indicate the standard error of the mean (SEM) values. *P < 0.05, ***P < 0 .001.
