Identification of host factors that promote SARS-CoV-2 infection of HEK293T-hACE2 cells. HEK293T-hACE2 cells transduced with the Brunello sgRNA library were infected with SARS-CoV-2 at MOI 0.01 and sgRNAs in resistant clones sequenced. Resistant clones were reinfected with SARS-CoV-2 at MOI 0.01 or MOI 0.1 and sgRNAs in resistant clones sequenced. For all three infections, MAGeCK analysis of multiple replicates compared to uninfected control library replicates yielded log2fold changes (log2FC) that were plotted on the x-axis. Negative log10-transformed FDR were plotted on the y-axis. Data are presented for the initial infection (A), MOI 0.01 reinfection (B), and MOI 0.1 reinfection (C).
